  • Resumo Summary

    A citricultura é uma das principais atividades agroindustriais do Estado de São Paulo. Atualmente, os custos de produção têm aumentado em decorrência do manejo necessário para a manutenção dos pomares contra o Citrus Greening (HLB), doença causada pela α-proteobacterium Candidatus Liberibacter asiaticus (CLas) e principal ameaça à citricultura mundial. Ter genótipos com maior tolerância à bactéria é um grande desafio e o uso da edição gênica poderia ser uma possível solução.

    Nesta proposta, pretende-se estabelecer um pipeline eficiente que permita a produção de mutações direcionadas em larga escala utilizando CRISPR/Cas9, para promover alterações em determinadas respostas de plantas cítricas quando infectadas com CLas. A análise da expressão gênica de genótipos suscetíveis, tolerantes e resistentes ao HLB após oito meses de infecção identificará genes que podem ajudar a explicar a interação entre alguns fenótipos característicos do HLB e genótipos de citros e gêneros relacionados, que são os alvos para o desenvolvimento desta proposta. A aplicação da tecnologia CRISPR no estudo do patossistema citros-HLB ampliará o conhecimento dos mecanismos envolvidos na resposta do HLB, permitindo o desenvolvimento de novas estratégias de combate à doença.

    A vaga está aberta a brasileiros e estrangeiros. O selecionado receberá Bolsa de Pós-Doutorado da FAPESP no valor de R$ 7.373,10 mensais e Reserva Técnica equivalente a 10% do valor anual da bolsa para atender a despesas imprevistas e diretamente relacionadas à atividade de pesquisa.

    Citriculture is one of the main agro-industrial activities in the state of São Paulo, Brazil. Currently, production costs have increased as a result of the management necessary for the maintenance of orchards against Citrus Greening (HLB), a disease caused by the α-proteobacterium Candidatus Liberibacter asiaticus (CLas) and the main threat for citriculture worldwide. Having genotypes with increased tolerance to the bacteria is a big challenge and the use of gene editing could be a possible solution.

    In this proposal, it is intended to establish an efficient pipeline that allow the production of targeted mutations on a large scale using CRISPR/Cas9, to promote changes in certain responses of citrus plants when infected with CLas. Gene expression analysis of susceptible, tolerant, and resistant genotypes to HLB after eight months of infection will identify genes that can help explain interaction between some phenotypes characteristic of HLB and citrus genotypes and related genera, which are the targets for development of this proposal. The application of CRISPR technology in the study of the citrus-HLB pathosystem will expand the knowledge of mechanisms involved in the HLB response, allowing the development of new strategies for fighting the disease.

    Studies will be carried out at the Citrus Biotechnology Laboratory at the Sylvio Moreira Advanced Citrus Research Center, affiliated with the Agronomic Institute (IAC) of São Paulo State Department of Agriculture and Supply

    This opportunity is open to candidates of any nationality. The selected candidate will receive a Post-Doctoral fellowship from the São Paulo Research Foundation (FAPESP) in the amount of R$ 7,373.10 monthly and a research contingency fund, equivalent to 10% of the annual value of the fellowship which should be spent in items directly related to the research activity.
