  • Resumo Summary

    O subprojeto "Estudo Estrutural e Funcional de Variantes da Hemoglobina Humana" (Pesquisadora Principal: Maria de Fatima Sonati) compõe o projeto temático "Doenças dos Glóbulos Vermelhos: Fisiopatologia e Novas Abordagens Terapêuticas" (Coordenador: Fernando Ferreira Costa). 

    Seu objetivo é, por meio de técnicas computacionais e experimentais, estudar alterações no alosterismo da hemoglobina humana que modifiquem suas propriedades funcionais, bem como sua estabilidade proteica, em variantes estruturais associadas a quadros clínicos e hematológicos relevantes.
     
    Pretende-se aqui ampliar e aprofundar estudos funcionais e estruturais de hemoglobinas humanas anômalas detectadas em nossa população, utilizando técnicas como a cristalização proteica, SAXS (espalhamento de Raios-X a baixos ângulos), espectroscopia RAMAN, Dicroismo Circular e, eventualmente, Microscopia Eletrônica de Alta Definição. 

    Do ponto de vista computacional, técnicas mais aprofundadas de análise de dinâmica molecular deverão ser aplicadas. Espera-se que, além de melhor caracterizar as variantes estruturais já em estudo, tal trabalho possibilite melhor compreensão de aspectos da dinâmica estrutural da Hb nativa por meio da perda, substituição ou inserção de resíduos que promovam diferentes respostas biofísicas. 

    As áreas de conhecimento envolvidas nesse subprojeto são, além da Hematologia, a Biofísica, a Bioquímica, a Biologia Molecular e Celular, as Ciências Farmacêuticas e a Patologia Clínica. 

    Os requisitos obrigatórios do candidato são a conclusão do Doutorado nos últimos cinco anos em uma das áreas supra citadas ou correlatas, o domínio da língua inglesa, experiência em análises estruturais de macromoléculas e com técnicas de biologia molecular e celular. 

    O requisito desejável é a realização de estágio prévio, com duração mínima de seis meses, em laboratório de instituição internacional de pesquisa no exterior. 

    Juntamente com o currículo, o candidato deve apresentar uma carta de interesse, com as justificativas e expectativas em relação ao pós-doutoramento no tema abrangido por este subprojeto, bem como duas cartas de recomendação de ex-professores ou ex-supervisores.

    The sub-project "Structural and Functional Study of Human Hemoglobin Variants" (Principal Investigator: Maria Fatima Sonati) is part of the thematic project "Diseases of Red Blood Cells: Pathophysiology and New Therapeutic Approaches" (Coordinator: Fernando Ferreira Costa). 

    Our goal is, through computational and experimental techniques, to study changes in the allosterism of the human hemoglobin that modify its functional properties as well as the protein stability, in structural variants associated with clinical and hematological important alterations. 

    It is intended to obtain broaden and deepen structural and functional studies of abnormal human hemoglobins detected in our population, using techniques such as protein crystallization, SAXS (X-ray scattering at low angles), Raman spectroscopy, Circular Dichroism and eventually Electron Microscopy High Definition. 

    From the computational point of view, more detailed techniques of molecular dynamics analysis must be applied. It is expected that, in addition to better characterize the structural variants being studied, such work enables better understanding of aspects of structural dynamics of native Hb through loss, substitution or insertion of residues that promote different biophysical responses. 

    The areas of knowledge involved in this subproject are Hematology, Biophysics, Biochemistry, Molecular and Cellular Biology, Pharmaceutical Sciences and Clinical Pathology. 

    The candidate's mandatory requirements are the completion of the doctorate in the last five years in one of the above mentioned or related areas, the knowledge of English, experience in structural analysis of macromolecules and molecular and cell biology techniques. 

    The desirable requirement is the completion of previous training, lasting at least six months, in a research laboratory of an international institution abroad.
     
    Along with the curriculum, the applicant must submit a letter of interest with the reasons and expectations for the post-doctorate in the subject covered by this subproject, as well as two letters of recommendation from former supervisors, to Profa. Maria de Fatima Sonati (sonati@fcm.unicamp.br).

    This opportunity is open to candidates of any nationalities. The selected candidate will receive a FAPESP's Post-Doctoral fellowship in the amount of R$ 6,143.40 (around 2,600 USD) monthly and a research contingency fund, equivalent to 15% of the annual value of the fellowship which should be spent in items directly related to the research activity.
