• Resumo Summary

  O projeto pretende investigar mecanismos de transformação tumoral em células deficientes em processos de reparo de DNA ou síntese translesão, notadamente xeroderma pigmentosum (XP), através de lesões induzidas no genoma por luz solar. Como modelo de estudo serão avaliadas células provenientes de pacientes afetados, irradiadas com luz em diferentes comprimentos de onda. Estamos particularmente interessados em avaliar os processos de oxidação na inibição de processos de reparo de DNA. Pretendemos identificar fenótipos de transformação e posterior avaliação dos níveis de mutagênese celular através de sequenciamento de nova geração (NGS).

  Impreterivelmente, os candidatos devem ter experiência prévia em cultura celular humana, e avaliação de fenótipo tumoral. Deve também apresentar conhecimento de biologia celular e molecular; experiência prévia em trabalhos em processos de reparo de DNA, detecção de lesão no DNA e avaliação de formação de radicais de oxigênio (ROS) nas células também serão consideradas.

  A vaga está aberta a brasileiros e estrangeiros. O selecionado receberá Bolsa de Pós-Doutorado da FAPESP no valor de R$ 7.373,10 mensais e Reserva Técnica equivalente a 10% do valor anual da bolsa para atender a despesas imprevistas e diretamente relacionadas à atividade de pesquisa.

  The project intends to investigate tumor transformation mechanisms in cells deficient in DNA repair or translesion synthesis processes, notably xeroderma pigmentosum (XP), caused by DNA damage by sunlight. As a study model, cells from affected patients will be evaluated, after irradiation with light of different wavelengths. We are particularly interested in mechanisms of oxidation of DNA repair proteins, with the inhibition of damage removal. The aim is the identification of possible novel transformation mechanisms. Subsequently, the candidate will assess mutagenesis levels through new generation sequencing (NGS).

  Candidates must have previous experience in human cell culture, and preferentially capacity to evaluate transformation phenotypes. Must also know cell and molecular biology; previous work on DNA repair processes, DNA damage detection, ROS detection and tumor transformation will also be considered. The fellow will be based in the Biomedical Sciences Institute of the University of São Paulo, Brazil.

  This opportunity is open to candidates of any nationalities. The selected candidate will receive a Post-Doctoral fellowship from the São Paulo Research Foundation (FAPESP) in the amount of R$ 7,373.10 monthly and a research contingency fund, equivalent to 10% of the annual value of the fellowship which should be spent in items directly related to the research activity.
