Resumo
Summary
O Centro de Pesquisa em Obesidade e Comorbidades (OCRC-IB/Unicamp) oferece uma bolsa de pós-doutorado para atuar na identificação e caracterização de proteínas associadas ao controle molecular da biogênese mitocondrial por meio da avaliação do interactoma funcional do co-repressor NCoR1.
Ainda, o candidato selecionado à bolsa estará engajado na identificação dos parceiros de interação do NCoR1 através de:
- Co-imunoprecipitação e espectrometria de massas do complexo;
- Investigação do papel do NCoR1 na biogênese mitocondrial durante a diferenciação de células musculares;
- Identificação de novos fatores de transcrição (TFs) que controlam a biogênese e função mitocondrial através da seleção de candidatos-alvo do interactoma usando repórteres biológicos;
- Investigação da interface NCoR1/TF como potencial alvo terapêutico em modelos in vitro de resistência à insulina.Os candidatos deverão ter conhecimento técnicos em imunoblot, qRT-PCR, co-imunoprecipitação, proteômica, imunoprecipitação de cromatina (ChIP), clonagem, e principalmente, ensaios de bioenergética mitocondrial. Além disso, é indispensável experiência prévia com cultura de células e manuseio de roedores (Rattus norvegicus e Mus musculus) bem como isolamento de células primárias de músculo desses animais.

b) Entrevista (pessoal ou via Skype).The Obesity and Comorbidities Research Center (OCRC-IB/UNICAMP) offers a post-doctoral fellowship to work in the identification and characterization of proteins associated with the molecular control of mitochondrial biogenesis through the functional interactome analysis of the co-repressor NCoR1.
The selected candidate will be involved in the identification of NCoR1 interaction partners through:
1) Co-immunoprecipitation and mass spectrometry experiments; investigation of NCoR1 role in mitochondrial biogenesis during muscle cell differentiation;
2) Identification of novel transcription factors (TFs) that control biogenesis and mitochondrial function through the selection of interactome target candidates using biological reporters; and
3) Investigation of the NCoR1/TF interface as a potential therapeutic target using in vitro models of insulin resistance.Applicants should be familiar with molecular biology techniques including imunoblot, qRT-PCR, co-immunoprecipitation, proteomics, chromatin immunoprecipitation (ChIP), cloning, and, importantly, mitochondrial bioenergetic assays. Besides that, is necessary previous experience with cell culture and rodents handling (Rattus norvegicus e Mus musculus) as well as isolation of primary skeletal muscle cells.

The selected candidate will receive a FAPESP’s Post-Doctoral fellowship in the amount of R$ 7,373.10 monthly and a research contingency fund, equivalent to 15% of the annual value of the fellowship which should be spent in items directly related to the research activity.
